Danijela \u0160anti\u0107, <\/strong>znanstvena savjetnica, Institut za oceanografiju i ribarstvo u Splitu i <strong>izv.prof. dr. sc. \u017deljka Trumbi\u0107, <\/strong>Sveu\u010dili\u0161ni odjel za studije mora Sveu\u010dili\u0161ta u Splitu.<\/p>\n\n\n\n<p><strong>Pozivaju se svi zainteresirani da prisustvuju obrani doktorskog rada.<\/strong><\/p>\n\n\n\n<p>Naslov: <strong>EKOLOGIJA MIKROBNE ZAJEDNICE S NAGLASKOM NA AEROBNE ANOKSIGENE FOTOTROFE U SREDNJEM JADRANU<\/strong><\/p>\n\n\n\n<p><strong>SA\u017dETAK<\/strong>:<\/p>\n\n\n\n<p>U ovom su radu tijekom jednogodi\u0161njeg istra\u017eivanja odre\u0111eni sastav i dinamika mikrobne zajednice na prostorno-sezonalnoj skali podru\u010dja srednjeg Jadrana s naglaskom na aerobne anoksigene fototrofe (AAF). Za odre\u0111ivanje strukture ukupne mikrobne zajednice na finijim taksonomskim razinama kori\u0161tena je kombinacija metabarkodiranja 16S rRNA gena te kvantitativnog CARD-FISH pristupa. Metabarkodiranjem <em>puf<\/em>M gena i IC-FISH metodom odre\u0111en je sastav zajednice AAF. Kako bi se otkrili obrasci unutar zajednica i protuma\u010dili u odnosu na najzna\u010dajnije okoli\u0161ne \u010dimbenike, provedeni su algoritmi umjetnih neuronskih mre\u017ea. Pripadnici klada SAR11 prevladavali su u raznolikim okoli\u0161nim uvjetima. Uo\u010den je op\u0107i obrazac pove\u0107anja ukupne mikrobne raznolikosti na ra\u010dun brojnosti u oligotrofnim uvjetima, posebice pove\u0107anjem saliniteta. Primije\u0107ena je zna\u010dajna sezonalnost u prosje\u010dnim apsolutnim AAF brojnostima (maksimumi u prolje\u0107e: 2,136 \u00b1 0,081 x 10<sup>4<\/sup>, minimumi ljeti: 0,86 x 10<sup>4<\/sup> stanica mL<sup>-1<\/sup>), IC-FISH grupama (klad <em>Roseobacter<\/em> prevalentan u jesen, razredi Alpha- i Gammaproteobacteria ljeti) i <em>puf<\/em>M podatcima (razlike u mjerama raznolikosti). Nisu uo\u010dene razlike u prostornom ni vertikalnom profilu. Zajednica je bila heterogenog sastava s najve\u0107im prosje\u010dnim relativnim udjelom klada <em>Roseobacter<\/em> (37,66%), a slijedili su pripadnici Gammaproteobacteria (35,25%) i op\u0107ih Alphaproteobacteria (31,15%). Otkrivena je prevalencija roda <em>Luminiphilus<\/em> na svim postajama, mjesecima i dubinama. Obrnuti odnos AAF brojnosti i mjera raznolikosti primije\u0107en je ljeti, ali nije uo\u010den za ostala godi\u0161nja doba. Visoka raznolikost zajednice bila je neovisna o temperaturi, salinitetu i trofi\u010dkom statusu. Dodatno je izvr\u0161ena detaljna usporedba pet metoda izolacije DNA i izravnog PCR-a na uzorcima \u201emock\u201c i morske zajednice kao temeljnog koraka u metabarkodiranju. Nijedna metoda nije bila najbli\u017ea teoretski o\u010dekivanom sastavu \u201emock\u201c zajednice. Dokazano je da se standardni protokol temeljen na toksi\u010dnom fenolu mo\u017ee zamijeniti kra\u0107im i sigurnijim alternativama temeljenima na talo\u017eenju izopropanolom i etanolom, dok su rezultati izravnog PCR-a pokazali prednosti smanjenog vremena rukovanja uzorcima.<\/p>\n\n\n\n<p>Title:&nbsp; <strong>MICROBIAL COMMUNITY ECOLOGY WITH AN EMPHASIS ON AEROBIC ANOXYGENIC PHOTOTROPHS IN THE CENTRAL ADRIATIC SEA<\/strong><\/p>\n\n\n\n<p><strong>ABSTRACT<\/strong>:<\/p>\n\n\n\n<p>In this thesis, composition and dynamics of the microbial community of the central Adriatic were determined during a one year study on a spatio-temporal scale, with an emphasis on aerobic anoxygenic phototrophs (AAPs). A combination of 16S rRNA metabarcoding and quantitative CARD-FISH approach was used to determine the structure of the total microbial community at finer taxonomic scale. The composition of the AAP community was estimated by <em>puf<\/em>M gene metabarcoding and IR-FISH method. In order to detect and interpret patterns within microbial communities in relation to the most significant environmental factors, artificial neural network algorithms were implemented. Members of SAR11 clade prevailed in diverse environmental conditions. A general pattern was recorded of total microbial diversity increase at the expense of abundance in oligotrophic conditions, especially with increased salinity. A significant seasonality was detected in average absolute AAP counts (maxima in spring: 2.136 \u00b1 0.081 x 10<sup>4 <\/sup>cells mL<sup>-1<\/sup>, minima in summer: 0.86 x 10<sup>4<\/sup> cells mL<sup>-1<\/sup>), IR-FISH groups (<em>Roseobacter<\/em> clade predominant in autumn, classes Alpha- and Gammaproteobacteria in summer) and <em>puf<\/em>M dataset (differences in diversity measures). No differences were observed in the spatial nor vertical profiles. The community had a heterogeneous composition with the highest average relative proportion of the <em>Roseobacter<\/em> clade (37.66%), followed by members of Gammaproteobacteria (35.25%) and Alphaproteobacteria (31.15%). The prevalence of genus <em>Luminiphilus<\/em> was revealed at all stations, months and depths. An inverse relationship between AAP abundance and diversity was observed in summer, but not for other seasons. High AAP diversity was independent of temperature, salinity and trophic status of the environment. Additionally, a detailed comparison of five DNA isolation protocols and direct PCR was performed on mock and marine community samples as a fundamental step in metabarcoding. No method was closest to the theoretically expected mock community composition.
