Larisa Zorani\u0107<\/strong>, Prirodoslovno-matemati\u010dki fakultet Sveu\u010dili\u0161ta u Splitu.<\/p>\n\n\n\n<p>Pozivaju se svi zainteresirani da prisustvuju javnoj obrani.<\/p>\n\n\n\n<p><strong>Naslov:<\/strong><\/p>\n\n\n\n<p><strong>Razli\u010diti aspekti mehanizma djelovanja antimikrobnih peptida kroz simulacijske studije<\/strong><\/p>\n\n\n\n<p><strong>Sa\u017eetak<\/strong>:<\/p>\n\n\n\n<p>Antimikrobni peptidi (AMP) su obe\u0107avaju\u0107a alternativa klasi\u010dnim antibioticima jer imaju \u0161iroko antimikrobno djelovanje, a\u202f patogeni na njih te\u017ee razvijaju otpornost.\u202f Osim prirodnih, sintetiziraju se novi AMP s ciljem ja\u010deg antimikrobnog djelovanja i smanjenja toksi\u010dnosti na ljudske stanice.\u202f\u202f U ovom radu simulacijama metodom molekularne dinamike istra\u017eivat \u0107e se\u202f mehanizam djelovanja i karakteristi\u010dna svojstva AMP-a: anisaxina, kiadina i adepantina, u interakcijama s modelima eukariotskih i bakterijskih membranama.\u202f Analizirat \u0107e se kako broj i polo\u017eaj aminokiseline Gly mijenja svojstva peptida i interakciju s membranom. Istra\u017eit \u0107e se djelovanje peptida s Gram-negativnom i Gram-pozitivnom membranom. Ispitat \u0107e se me\u0111ukorak u mehanizmu djelovanja koji uklju\u010duje sferi\u010dna ispup\u010denja gornjeg dijela lipidnog dvosloja, opa\u017een pri velikim koncentracijama peptida na povr\u0161ini membrane, te je li takva deformacija mo\u017ee uzrokovati ekstrakciju lipida. Cilj je bolje razumijevanje mehanizma djelovanja i\u202f otkrivanje potencijalnog antibiotskog djelovanja.<\/p>\n\n\n\n<p><strong>Title:<\/strong><\/p>\n\n\n\n<p><strong>Different aspects of the mechanism of actions of antimicrobial peptides throughout simulation studies<\/strong><\/p>\n\n\n\n<p><strong>Abstract<\/strong>:<\/p>\n\n\n\n<p>Antimicrobial peptides (AMPs) are a promising alternative to conventional antibiotics due to their broad spectrum of antimicrobial activity, to which pathogens have difficulty developing resistance. In addition to natural ones, new AMPs are being synthesized to achieve more potent antimicrobial activity and reduce toxicity to human cells. In this thesis, using molecular dynamics simulations, we will investigate the interaction of the following AMPs: anisaxin, kiadin and adepantin with eukaryotic and bacterial membrane models to determine their characteristic properties and mechanism of actions. We will analyze the influence of the number and positions of amino acid Gly in the primary sequence on the peptide activity. Also, we will explore differences in the interaction of AMPs with Gram-negative and Gram-positive membranes. We will test if spherical bulges of the upper bilayer leaflet, observed at high peptide concentrations on the membrane surface, are part of the mode of action of AMPs, and whether such deformation can cause lipid extraction.
