Marija Ragu\u017e<\/strong>, Medicinski fakultet Sveu\u010dili\u0161ta u Splitu.<\/p>\n\n\n\n<p>Pozivaju se svi zainteresirani da prisustvuju javnoj obrani.<\/p>\n\n\n\n<p><strong>Naslov:<\/strong><\/p>\n\n\n\n<p><strong>Optimizacija elektroformacije divovskih unilamelarnih vezikula za razli\u010dite lipidne smjese uz naglasak na visoke koncentracije kolesterola<\/strong><\/p>\n\n\n\n<p><strong>Sa\u017eetak<\/strong>:<\/p>\n\n\n\n<p>Divovske unilamelarne vezikule (DUV) \u010desto su kori\u0161teni modeli stani\u010dnih membrana. Najra\u0161irenija metoda proizvodnje DUV-ova je elektroformacija. Prvi korak metode nano\u0161enje je tankog lipidnog filma na elektrodu. Na film se potom nanosi vodena otopina te se primjenjuje elektri\u010dno polje kako bi ga se odvojilo od povr\u0161ine i stvorilo DUV-ove. Iako jednostavna na prvi pogled, metoda ovisi o puno parametara te ju je potrebno optimizirati ovisno o lipidnom sastavu kako bi se dobili DUV-ovi visoke kvalitete za daljnje eksperimente. Poseban naglasak na\u0161eg istra\u017eivanja bit \u0107e na membranama s visokim koncentracijama kolesterola koje dose\u017eu i prelaze prag saturacije u membranskom dvosloju. Ovakvi sustavi potrebni su za istra\u017eivanje uloge kolesterola u membranama vlaknastih stanica le\u0107e oka i razumijevanje procesa&nbsp; nastanka ateroskleroze. Ve\u0107ina stani\u010dnih membrana sadr\u017ei ni\u017ee koncentracije&nbsp; kolesterola i dosada\u0161nji protokoli za takve smjese daju vezikule zadovoljavaju\u0107e kvalitete. Sistemati\u010dna optimizacija elektroformacije za sustave s visokom&nbsp; koncentracijom kolesterola nikada nije provedena. Posljedi\u010dno, u istra\u017eivanju \u0107emo se fokusirati na prilagodbu elektroformacije DUV-ova kori\u0161tenjem razli\u010ditih lipidnih&nbsp; sastava s visokim koncentracijama kolesterola. Tako\u0111er, identificirat \u0107emo mogu\u0107e prepreke i nedostatke te ponuditi potencijalna rje\u0161enja.<\/p>\n\n\n\n<p><strong>Title:<\/strong><\/p>\n\n\n\n<p><strong>Optimization of giant unilamellar vesicle electroformation for different lipid mixtures with focus on high cholesterol concentrations<\/strong><\/p>\n\n\n\n<p><strong>Abstract<\/strong>:<\/p>\n\n\n\n<p>Giant unilamellar vesicles (GUVs) are widely used as artificial cell membrane models. Electroformation is the most commonly used method for their production. The first step is the deposition of a thin lipid film on an electrode. The film is then hydrated and an alternating current applied in order to detach the film and form GUVs. Although simple at first glance, the method depends on multiple parameters, so optimization is required for every new mixture used in order to obtain high quality GUVs for further experiments. Our research will focus on membranes with cholesterol concentrations reaching and surpassing the bilayer saturation threshold. These model membranes are needed for studies of the role of cholesterol in the fiber cell plasma membranes of the eye lens or development of atherosclerosis. Most cell membranes contain much lower cholesterol concentrations, and protocols developed so far have yielded sufficiently good vesicles for such mixtures. However, the electroformation method has not yet been optimized for high cholesterol content mixtures. Consequently, our research will focus on optimization of GUVs electroformation using different lipid mixtures and high cholesterol concentrations.
