Roko Andri\u010devi\u0107<\/strong> \u2013 Fakultet gra\u0111evinarstva, arhitekture i geodezije Sveu\u010dili\u0161ta u Splitu.<\/p>\n\n\n\n<p>Pozivaju se svi zainteresirani da prisustvuju javnoj obrani.<\/p>\n\n\n\n<p><strong>Naslov:<\/strong><\/p>\n\n\n\n<p><strong>Utjecaj podmorskih ispusta pro\u010di\u0161\u0107ene otpadne vode na strukturu mikrobne zajednice i rezistom priobalnog mora<\/strong><\/p>\n\n\n\n<p><strong>Sa\u017eetak<\/strong>:<\/p>\n\n\n\n<p>Ve\u0107ina obalnih gradova koristi podmorske kanalizacijske ispuste za odlaganje pro\u010di\u0161\u0107enih otpadnih voda u okoli\u0161, me\u0111utim, njihov utjecaj na morski ekosustav je vrlo kompleksan i jo\u0161 uvijek nedovoljno istra\u017een. Standardna procjena kakvo\u0107e morske vode uklju\u010duje odre\u0111ivanje broja indikatorskih bakterija, dok je primjenom razli\u010ditih selektivnih podloga i tehnika mogu\u0107e uzgojiti manje od 1% poznatih bakterijskih vrsta. Razvoj metagenomike omogu\u0107io je uvid u cjelokupan sastav mikrobne zajednice kao i mogu\u0107nost vrlo detaljnih istra\u017eivanja pojedinog njezinog dijela ili karakteristike, poput okoli\u0161nog rezistoma i uloge okoli\u0161a u razvoju i \u0161irenju globalnog problema antibiotske rezistencije. Stoga, glavni fokus ove disertacije je primjenom mikrobiolo\u0161kih, molekularno-geneti\u010dkih te bioinformati\u010dkih analiza istra\u017eiti utjecaj podmorskih kanalizacijskih ispusta na strukturu mikrobne zajednice u obalnim vodama srednjeg Jadrana, s posebnim naglaskom na unos klini\u010dki zna\u010dajnih patogenih bakterija i njihovih gena antibiotske rezistencije, kao i procijeniti \u010dimbenike okoli\u0161a koji tome doprinose. Nakon vi\u0161emjese\u010dnih terenskih mjerenja i uzorkovanja na lokacijama dvaju podmorskih ispusta, analizirat \u0107e se struktura mikrobioma i rezistoma, utvrditi u\u010destalost pojave antibiotske rezistencije te istra\u017eiti fenotip i genotip rezistencije odabranih patogenih bakterija. Dobiveni rezultati \u0107e pru\u017eiti nove spoznaje o podmorskim ispustima kao epidemiolo\u0161ki va\u017enim izvorima ali i \u017eari\u0161tima one\u010di\u0161\u0107enja okoli\u0161a rezistentnim bakterijama i genima antibiotske rezistencije. Osim toga, rezultati ovog rada doprinijet \u0107e razvoju obuhvatnijih strategija u cilju u\u010dinkovitije kontrole i smanjenja one\u010di\u0161\u0107enja obalnog morskog okoli\u0161a te pobolj\u0161anju za\u0161tite zdravlja ljudi na lokalnoj i globalnoj razini.<\/p>\n\n\n\n<p><strong>Title:<\/strong><\/p>\n\n\n\n<p><strong>The impact of submarine outfalls of treated wastewater on the microbial community structure and the resistome of the coastal marine environment<\/strong><\/p>\n\n\n\n<p><strong>Abstract<\/strong>:<\/p>\n\n\n\n<p>Most coastal cities use submarine outfalls to discharge treated wastewater into the environment, but their impact on the marine ecosystem is very complex and not well understood. Standard assessment of seawater quality involves enumeration of indicator bacteria, while less then 1% of known bacterial species can be cultured using various selective media and techniques. The development of metagenomics has enabled insight into the overall structure of the microbial community as well as the possibility of very detailed study of its individual parts or characteristics, such as the environmental resistome and the role of the natural environment in the development and spread of the global problem of antibiotic resistance. Therefore, the main focus of this dissertation is to study the impact of submarine sewage discharges on the microbial community structure in the coastal waters of the central Adriatic Sea, with emphasis on the introduction of clinically significant pathogenic bacteria and their antibiotic resistance genes, using microbiological, molecular genetic and bioinformatics analyses, as well as to assess the environmental factors promoting such changes. Following montly field measurements and sampling at the sites of two submarine outfalls, the structure of the microbiome and associated resistome will be analysed, the frequency of antibiotic resistance will be determined, and the phenotype and genotype of antibiotic resistance of selected pathogenic bacteria will be investigated. The obtained results will provide new insights about the submarine outfalls as epidemiologically important sources as well as hotspots of environmental pollution with antibiotic-resistant bacteria and associated resistance genes.
