Larisa Zorani\u0107<\/strong> \u2013 Prirodoslovno-matemati\u010dki fakultet Sveu\u010dili\u0161ta u Splitu.<\/p>\n\n\n\n<p>Pozivaju se svi zainteresirani da prisustvuju javnoj obrani.<\/p>\n\n\n\n<p>Naslov:<\/p>\n\n\n\n<p><strong>Razli\u010diti aspekti mehanizma djelovanja antimikrobnih peptida kroz simulacijske studije<\/strong><\/p>\n\n\n\n<p><strong>Sa\u017eetak<\/strong>:<\/p>\n\n\n\n<p>Metodom molekulske dinamike istra\u017eeni su mehanizmi djelovanja i karakteristi\u010dna svojstva triju vrsta antimikrobnih peptida (AMP-a) (adepanina-1, diPGLa-H te \u0161est njegovih analoga kiadina i anisaxina-2S) u interakciji s modelima plazma membrana Gram-pozitivnih, unutarnjih i vanjskih membrana Gram-negativnih bakterija, te neutralnih membrana eukariota. Izvedene su atomisti\u010dke, zdru\u017eene atomske i krupnozrnate simulacije. Simulacije istra\u017euju va\u017enost strukturiranja peptida, njihove fleksibilnosti, udru\u017eivanja u klastere i drugih svojstava va\u017enih za antimikrobno djelovanje. Pokazano je kako asocijacije peptida mogu pomo\u0107i postavljanju peptida u polo\u017eaj i konformaciju povoljnu za njihovo umetanje u membranu. Analiza kako&nbsp; broj i polo\u017eaj aminokiseline glicin mijenja svojstva peptida i utje\u010de na njegovu interakciju s membranom pokazala je da je va\u017eniji polo\u017eaj, nego sam broj glicina u sekvenci peptida. Potvr\u0111eno je da mehanizam djelovanja AMP-a mo\u017ee uklju\u010divati potpunu ekstrakciju lipida iz gornjeg membranskog sloja ostvarenu uz velike lokalne koncentracije peptida na povr\u0161ini membrane. Mogu\u0107e je da je takva ekstrakcija lipida prvi korak u mehanizmu djelovanja i da rezultiraju\u0107a asimetrija u lokalnoj gusto\u0107i lipida u dva membranska sloja mo\u017ee smanjiti energetsku barijeru za umetanje peptida. Ovaj rad doprinosi boljem razumijevanju mehanizma djelovanja i otkrivanju antibiotskog potencijala antimikrobnih peptida.<\/p>\n\n\n\n<p>Title:<\/p>\n\n\n\n<p><strong>Different aspects of the mechanism of actions of antimicrobial peptides throughout simulation studies<\/strong><\/p>\n\n\n\n<p><strong>Abstract<\/strong>:<\/p>\n\n\n\n<p>Using the molecular dynamics method, the mechanisms of action and characteristic properties of three types of antimicrobial peptides (AMPs) (adepanin-1, diPGLa-H and six of its analogues kiadins and anisaxin-2S) were investigated in interaction with plasma membrane models of Gram-positive, inner and outer membranes of Gram-negative bacteria, and neutral membranes of eukaryotes. Atomistic, united atom and coarse-grained simulations were performed. The simulations investigate the importance of peptide structuring, their flexibility, clustering and other properties important for antimicrobial activity. It is shown how peptide associations can help to place peptides in a position and conformation favorable for their insertion into the membrane. Analysis of how the number and position of the amino acid glycine changes the properties of the peptide and affects its interaction with the membrane showed that the position is more important than the number of glycine in the peptide sequence. It has been confirmed that the mechanism of action of AMP can include the complete extraction of lipids from the upper membrane layer achieved with high local concentrations of peptides on the membrane surface. It is possible that this type of lipid extraction is the first step in the mechanism of action, and that the resulting asymmetry in the local density of lipids in the two membrane layers can reduce the energy barrier for peptide insertion.
